Set up THP????

For regulation THP is consiglabile to effect him/it with turned on car or out?
I ask this because there are some due variations to the tension of the battery, for instance if I measure THP with the out car THP=22, with turned on car THP=23 ,8.
Can this variation be meaning? Especially THP hi can create you lack me?
